Alkylation of Intracellular and Extracellular DNA by Dimethylnitrosamine following Activation by Isolated Rat Hepatocytes1
نویسندگان
چکیده
Freshly prepared rat hepatocytes isolated by perfusion with collagenase were able to metabolize p.M concentrations of dimethylnitrosamine to a methylating agent. The methylation of hepatocyte DNA in this system was complete within 2 hr, and after this time, the content of O6-methylguanine in the DNA declined, showing that the repair system for this product was active in the isolated hepatocytes. When extracellular calf thymus DNA was added to the incubated hepatocytes, this also became methylated. Methylation of this DNA was not due to cell lysis releasing activating enzymes into the medium, show ing that the methylating species formed by the hepatocytes from dimethylnitrosamine is sufficiently stable to pass out of the cell in substantial amounts. These results support the possibility that alkylation of liver cells would not be confined to those cells metabolizing dimethylnitrosamine but could be ex tended to those cells which are in close proximity to the activating cells. These cells could include nonparenchymal cells which are known to be targets for the carcinogenic action of dimethylnitrosamine.

1. N[14C]-Methyl-N-nitrosourea, [14C]dimethylnitrosamine, [14C]dimethyl sulphate and [14C]methyl methanesulphonate were injected into rats, and nucleic acids were isolated from several organs after various time-intervals. Radioactivity was detected in DNA and RNA, partly in major base components and partly as the methylated base, 7-methylguanine. 2. No 7-methylguanine was detected in liver DNA ...

The effects of phénobarbital(PB) and other liver monooxygenase modifiers on dimethylnitrosamine (DMN)-induced alkylation of rat liver DNA and protein were investigated at different carcin ogen doses. In rats given single injections of radioactively labeled DMN, pretreatment with PB (80 mg/kg body weight, adminis tered for 5 days) resulted in a small but significant decrease in the formation of...

Dacarbazine (DTIC) is a synthetic chemical antitumor agent which is used to treat malignant melanoma and Hodgkin’s disease. DTIC is a prodrug which is converted to an active form undergoing demethylation by liver enzymes. The active form prevents the progress of disease via alkylation of DNA strand. In the structure of this drug, the imidazole ring, a triazen chain and carboxamide group ex...
